TurretGirls puts you in control of a turreted operative who fights off alien attacks from unstable wormholes. The main mission is to protect the generator, the sole source of power for a planetary superweapon. Operating from a fixed rail platform, the heroine confronts swarms of alien invaders that threaten the entire installation. The survival of the planet depends on this lone defense against the invading horde, as retreat is impossible.

Success in TurretGirls depends on the gamer's combat effectiveness, as each completed mission brings resources for upgrades. The funds received are used to strengthen defenses, build frontline generators and increase overall firepower. The rail turret platform itself receives systemic upgrades based on results in combat. High points also unlock elite accessories and various underwear, serving as tangible rewards for successful actions against the alien threat.

The protagonist is a member of the Tactical Strike Unit for the Neutralization of Planetary Aliens and wears a specialized uniform. This gear is designed to be strategically destructible in extreme battle conditions. During intense clashes, the suit self-destructs, which is a function to increase the heroine's survivability and maneuverability. This feature puts mission success and combat effectiveness above the structural integrity of the outfit.
